Механизмы клеточного старения, индуцированного сочетанным действием физических и химических факторов в концентрациях, близких к предельно допустимым или незначительно их превышающих, остаются недостаточно изученными. Наиболее информативным методом верификации данных причинно-следственных связей выступают экспериментальные исследования на модельных организмах.</p><p>Цель исследования — выявить и экспериментально оценить комплекс биомаркеров, ассоциированных с ускоренным клеточным старением и развитием органных повреждений, у белых крыс при длительном изолированном, комбинированном и сочетанном воздействии физических и химических факторов производственной среды на уровне 1,5 ПДК/ПДУ.</p><p>Проведён 180-суточный эксперимент на 180 крысах линии Wistar. Животные были разделены на 6 групп: воздействие общей вибрации; шума; химическое воздействие (смесь ароматических углеводородов на уровне 1,5 предельно допустимой концентрации); комбинированное физическое воздействие (вибрация + шум); сочетанное физико-химическое воздействие (вибрация + шум + смесь углеводородов); интактный контроль. Дополнительно в фоновых исследованиях использовано 10 особей. Оценивали массу тела, поведение в тесте «открытое поле». На 60-е, 120-е и 180-е сутки проводили биохимический и клинический анализ крови и мочи, гистологическое исследование внутренних органов, определяли относительную длину теломер в мышечной ткани методом полимеразной цепной реакции в реальном времени. Статистический анализ проводили с использованием непараметрических критериев (Манна–Уитни).</p><p>К 180-м суткам эксперимента у животных опытных групп зарегистрирован симптомокомплекс ускоренного старения: снижение поведенческой активности, метаболические нарушения (дислипидемия, гипергликемия), дисфункция печени и почек, полиорганные структурные повреждения. Установлено достоверное укорочение теломер во всех опытных группах, наиболее выраженное при комбинированном и сочетанном воздействии.</p><sec><title>Ограничения исследования</title><p>Ограничения исследования. Исследование проводилось на модели грызунов; прямые корреляции с человеческой популяцией требуют валидации в эпидемиологических исследованиях.</p></sec><sec><title>Выводы</title><p>Выводы. Длительное воздействие вибрации, шума и химических веществ индуцирует у крыс состояние хронического стресса с формированием комплекса биомаркеров ускоренного клеточного старения: поведенческих дефицитов, метаболических нарушений, полиорганной дисфункции и укорочения теломер.</p></sec><sec><title>Этика</title><p>Этика. Исследование выполнено в соответствии с этическими принципами работы с лабораторными животными (Европейская конвенция о защите позвоночных животных). The mechanisms of cellular aging induced by the combined action of physical and chemical factors at concentrations close to or slightly exceeding the maximum permissible levels remain poorly understood. Experimental studies on model organisms are the most informative method of verifying these causal relationships.</p><p>The study aims to identify and experimentally evaluate a complex of biomarkers associated with accelerated cellular aging and the development of organ damage in white rats with prolonged isolated, combined and combined exposure to physical and chemical factors of the industrial environment at the level of 1.5 MPC/MPL.</p><p>The authors have conducted a 180-day experiment on 180 Wistar rats. The experts have divided the animals into 6 groups: exposure to general vibration; noise; chemical exposure (a mixture of aromatic hydrocarbons at 1.5 maximum permissible concentration); combined physical exposure (vibration + noise); combined physico-chemical exposure (vibration + noise + mixture of hydrocarbons); intact control. Additionally, 10 individuals were used in background studies. The authors assessed body weight and behavior in the "open field" test. On days 60, 120 and 180 they performed biochemical and clinical blood and urine tests, as well as a histological examination of internal organs, the scientists determined the relative length of telomeres in muscle tissue using a polymerase chain reaction in real time. They performed statistical analysis using nonparametric criteria (Mann–Whitney).</p><p>By the 180th day of the experiment, the animals of the experimental groups showed a symptom complex of accelerated aging: decreased behavioral activity, metabolic disorders (dyslipidemia, hyperglycemia), liver and kidney dysfunction, and multiple organ structural damage. Significant telomere shortening was found in all experimental groups, most pronounced with combined and combined exposure.</p><sec><title>Limitations</title><p>Limitations. The study was conducted on a rodent model; direct correlations with the human population require validation in epidemiological studies.</p></sec><sec><title>Conclusion</title><p>Conclusion. Prolonged exposure to vibration, noise, and chemicals induces a state of chronic stress in rats with the formation of a complex of biomarkers of accelerated cellular aging: behavioral deficits, metabolic disorders, multiple organ dysfunction, and telomere shortening.</p></sec><sec><title>Ethics</title><p>Ethics.
