Работа в отрасли черной металлургии сопряжена с тяжёлыми условиями труда, которые могут повышать риск развития у рабочих хронических заболеваний, в том числе ожирения. Одним из механизмов действия вредных веществ, содержащихся в воздухе рабочей зоны, на организм является повышенное образование свободных радикалов, приводящих к окислительному стрессу. В данной работе проводили исследование влияния полиморфизмов генов CAT (rs1001179 и rs7943316) и GPX4 (rs713041), которые кодируют ферменты антиоксидантной системы, на индекс массы тела у работников предприятия черной металлургии.</p><p>Цель исследования — оценить генетический риск развития ожирения у сотрудников металлургического комбината.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Обследованы мужчины-работники конвертерного цеха (151 человек) и административно-управленческий персонал (203 человека). Выделение ДНК осуществляли набором LumiPure. Для определения полиморфизмов генов применяли коммерческие наборы и система Applied Biosystems QuantStudio 3. Для анализа результатов применяли пакет программ STATISTICA (критерии Манна–Уитни и Краскела–Уоллиса). Применялась кодоминантная и аллельная генетические модели наследования.</p></sec><sec><title>Результаты</title><p>Результаты. в основной группе обнаружены статистически достоверные различия. У работников конвертерного цеха с мутантным аллелем T по полиморфизму rs7943316 гена CAT наблюдались более высокие значения ИМТ (p=0,024). Аналогично, у рабочих в основной группе с мутантным аллелем T и мутантным гомозиготным генотипом TT гена GPX4 значения ИМТ были статистически выше (p=0,01 и p=0,015 соответственно).</p></sec><sec><title>Ограничения исследования</title><p>Ограничения исследования. Не определены количественные характеристики профессиональных вредных факторов.</p></sec><sec><title>Заключение</title><p>Заключение. Полиморфизмы rs7943316 гена CAT и rs713041 гена GPX4 снижают антиоксидантную защиту и могут быть одним из факторов риска развития ожирения у работников конвертерного цеха предприятия черной металлургии.</p></sec><sec><title>Этика</title><p>Этика. Этическое одобрение исследования было получено от локального этического комитета Федерального бюджетного учреждения науки «Екатеринбургский медицинский научный центр профилактики и охраны здоровья рабочих промышленных предприятий» Роспотребнадзора (протокол № 1 от 26.02.2021). Working in the ferrous metallurgy industry is associated with harsh working conditions, which can increase the risk of workers developing chronic diseases, including obesity. One of the mechanisms of action of harmful substances contained in the air of the working area on the body is the increased formation of free radicals, leading to oxidative stress. In this work, we studied the effect of polymorphisms of the CAT (rs1001179 and rs7943316) and GPX4 (rs713041) genes, which encode enzymes of the antioxidant system, on body mass index in employees of the ferrous metallurgy enterprise.</p><p>The study aims to assess the genetic risk of obesity among employees of the metallurgical plant.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The authors have examined male employees of the converter shop (151 people) and administrative and managerial staff (203 people). DNA isolation was performed with a LumiPure kit. Commercial kits and the Applied Biosystems QuantStudio 3 system were used to determine gene polymorphisms. To analyze the results, we used the STATISTICA software package (Mann–Whitney and Kraskel–Wallis criteria). Codominant and allelic genetic models of inheritance were used.</p></sec><sec><title>Results</title><p>Results. We found statistically significant differences in the main group. Converter shop workers with a mutant T allele for the rs7943316 polymorphism of the CAT gene had higher BMI values (p=0.024). Similarly, the workers in the main group with the mutant T allele and the mutant homozygous TT genotype of the GPX4 gene had statistically higher BMI values (p=0.01 and p=0.015, respectively).</p></sec><sec><title>Limitations</title><p>Limitations. The main occupational hazards have not been identified.</p></sec><sec><title>Conclusion</title><p>Conclusion. Polymorphisms rs7943316 of the CAT gene and rs713041 of the GPX4 gene reduce antioxidant protection and may be one of the risk factors for obesity in workers of the converter shop of the ferrous metallurgy enterprise.</p></sec><sec><title>Ethics</title><p>Ethics.
